If you just reinstalled the OS, it wouldn't know about the stuff you'd installed on E:\, you'd need to reinstall those in order for the registry etc. to know about them.
Additionally I'd like to know if it is possible to move the current boot
partition to another drive and dual boot it with the newly installed OS
until things are set up the way I like and then delete it without a trace,
so the system no longer dual boots?
Why not just take an image of your OS drive once you're happy with it? Then you can just restore that should you find you need to, and provided your E:\ drive remains the same (or just image that too) then you'll be exactly back at a starting point you like.
