I don't think that a queen would have been acceptable in those days.
Matilda was the last and there was civil war over her. At the time of
the alleged murder of the two princes, the Tudors weren't in any
position of power and influence, that came later, indeed the only
person to gain full advantage was Richard. That he engineered their
removal is an absotute certainty as far as I'm concerned.

A female with a legitimate claim would have become the puppet of one or
another of the Lords - not by-passed. It's not actually known when the
princes died - it could as easily have been the first act of Henry's reign,
and a lot of spin afterwards.
When Richard came to the throne, all of the Plantagenet heirs, including all
Edward's children except the 2 princes, and their mother were free and
taking pensions from him.

As to Henry (which-either?) getting rid of potential Plantagenet
heirs, there were far too many, and the only ones he did see off were
those that posed more than a possible threat. The execution of those
who plot against the sovereign wasn't started by Henry and didn't end
with him. Kingship was a very messy and dangerous thing in those days.

To marry Elizabeth, he needed her to be legitimate - but if she was legit,
then so were her brothers... Even though he fought his way to the throne
(killing a lot more than 2 boys one the way), leaving legit heirs about the
landscape asked for people to rebel.
