What you want to do is to relocate you iTunes library from the old
80GB drive to the new 500GB drive. I did this last year when I got a
new, larger external drive. It will take awhile to copy the files
over to the new drive. Once the copy completes and you can bring up
iTunes and make sure it's pointing to the new location, you can unplug
the old drive. When you sync the iPod, it will sync up wherever the
current library is located.
Look in iTunes help under "move itunes library". Or, you can google
the same. Here's one reference:

I don't think its quite this easy - if it was - everyone would be
copying their itunes library all over the place....and if you read the
lifehacker page - it says this can turn into a disaster.
I suggest you spend the $20 for "TouchCopy". I just purchased a new
laptop and used this program to move my iTunes library. There was no
way I was using UltraEdit to edit the ,xml file in itunes to redirect
where the library is located. "TouchPad" worked fine - I only got a
few errors - and one frozen iPod - but it all moved over ok to the new
machine. And my 2nd generation iPod actually still works. Of course
- now I need to get a new iPod to go with the new laptop.....:)

Seems to me that I could, for lack of a simpler solution and by investing
a
couple of hours of my time, restore the iPod to virgin state, uninstall
the
iTunes program, unplug the drive I no longer plan to use, reinstall the
iTunes and let it recreate the library with the music on the new drive.
The
big downside to that, of course, is that I'd have to recreate my
favorites
lists, and that's a big downside.
I will check out the TouchCopy website though.
Does your iTunes preference for "Music Folder Location" currently point to
the 80GB drive?
No. The iTunes preference for "Music Folder Location" points to
C:\Documents and Settings\Owner\My Documents\My Music\iTunes\iTunes Music.
But here's the deal. 90% of the music (in the music library and on the
iPod) is on the 80GB drive. The only music that is in the above folder is
the 10% that was converted from WMA to mp4 by iTunes.
Knowing that, do you have any other thoughts on this? Thanks.
Here's what I *thnk* I'd do if I were you (but **first** I'd read the Moving Your Library threads
over at iPodlounge.com forums - don't blame me if I miss some gotchas here!)
- Uncheck the "Keep iTunes Music Folder Organized" and "Copy files to iTunes Music Folder
When Adding to LIbrary
- change your Music Folder Location preference to point to the 500GB volume
- Check those first two preferences
- Choose the "Consolidate Library" menu item
I *think* that sequence will systematically copy all of your music off the two previous dscs into
the specified folder on the new disc and update your library files to reflect that. But again, read the
threads before you do it - it should all be covered.
