I think the answer is "No" in both cases, but let me see if I can find some chapter & verse on that.
Staves & wands are always fully charged when created; see DMG pg. 287.
As for ammunition, there's nothing in the DMG or the ROTG articles, but my suggestion would be to allow it but not give a break on the magic part of the price, e.g. if you only want 5 arrows instead of 50 they'd only pay 2030 GP instead of 2350 GP, since a single masterwork arrow costs 6 GP (PHB, pg. 122).
