I realize there is no definitive answer to this question. However, my vote
is Moneymaker's 88 against Brenes' AA mid way through the 2003 WSOP main
event. Moneymaker went all-in and Brenes called. An 8 came on the turn,
Moneymaker made a set, cracked Brenes' AA, and knocked him out of the
tournament. Most knowledgeable players don't dispute the impact of
Moneymaker's main event win. In fact, many believe this was the start of
the entire poker boom. If Moneymaker misses that 23:1 set on the turn, he's
finished.

I'd be interested in hearing any other nominations for "Most important hand
in poker history".

Al Bass dealt in 7 Card Stud two Royal Flushes in the same hand at the
Sahara cardroom in Vegas, I guessing back in the early 70's. RazzO
would know more about it. At a time when most of poker was still
being played in boiler rooms and such it got national and world wide
news coverage. People would come into the Sahara cardroom for years
afterwards wanting to know which table it happened on.

Jack Strauss' famous "chip and a chair" hand. Back in '82 Jack
silently pushed all his chips into the middle and was called chip for
chip by another player who ended up winning the pot. Jack thinking he
was out was told he still had a $500 which was found hidden under his
cocktail napkin. He was ruled still in the tournament since he hadn't
announced that he was all-in and the guy had only called. Jack went
on from that point to win the tournament and become the 1982 WSOP
champion.
