If there's a puncture, I usually try to fix it by patching without
removing the wheel from the bike (*). Once that fails and I have to
remove the wheel from the bike, I usually change the tube against the
spare. The broken tube gets patched right away, so that I have a working
spare tube for a second puncture (never happened yet).
With patching I do have a 2-out-of-3 success ratio - last fall I failed
to fix a tube - it had rolled in the tire while deflating and had
multiple secondary holes... When trying to find them, a seam ruptured...
(*) Lay the bike on the side, try to find the puncture. Remove one side
of the tire from the wheel for about 1/3rd of the circumference and get
the tube out. Fix as usual and re-mount. That's the easiest way to fix a
flat, especially if you have racks, trailer hitch, derailleur cage and
other stuff mounted on or around the axle...
Actually on trips around town I don't carry any repair kit except a
multi-tool and some (human-skin) patches. Nothing to repair a flat...
Depends on where you go and on how many spares vs. repair kit(s) you
want to carry...
