DVD Player on 10.5.1 on my MacBook Pro is fine with opening a DVD folder
copied to my hard drive.

The critical point: the DVD image must have a VIDEO_TS folder, and DVD
player effectively opens the parent folder (you can select either the
parent folder or VIDEO_TS). I don't know if it bothers checking for
AUDIO_TS but I have one in my test cases.

I have Quicktime Pro,Perian and the Quicktime MPEG2 component
installed. If I mount an ISO,pointing Quicktime to the Disk Image on
the desktop WILL play the DVD,but pointing Quicktime to the VIDEO_TS
folder within gives the 'unsupported media' message.

(I assume that you meant "DVD Player" instead of "QuickTime" in that
last sentence.)

I found a small and unprotected DVD that I could experiment with. I was
able to play it using DVD Player with every permutation I could easily
achieve. Where appropriate, I was able to play it due to DVD Player
auto-launching on DVD insertion or a disc being mounted, or in all cases
by using Open Media and navigating to the VIDEO_TS folder.

- Normal playback of the DVD.
- Copied the files to a folder on my hard drive.
- Created a read only disc image using Disk Utility (Mac OS Extended
file system).
- Created a toast disc image usign Toast (UDF file system).
- Created a bin/cue disc image using Toast (UDF file system).

I don't have any tools handy which can create a .iso file.

My earlier tests were playing across a network from a folder shared by
another Mac (AFP over Gigabit Ethernet).

Everything works fine, so the problem is not a general issue with DVD
Player on 10.5.1. It must be something specific to your computer, or the
format you have stored your images in. Can you try copying the files out
of one of the ISOs into a normal folder on your hard drive?

No other explanations occur to me at present.
