Check "Utilities" in "Applications" folder, you will see "Audio MIDI setup", run it and make sure output is at least 44.1 khz/16bit 2 channels (CD standard). It looks like sample rate problem to me.

Do you have any third party utility,plugin that deals with iTunes sound? Make sure to uninstall them until developers ship a iTunes 7 compatible version. For example, it refused to play here with OSS3D installed as plugin.

If you manually downloaded files, you may try re-installing, first Quicktime and after reboot, iTunes.

I see you didn't get any reply so I thought I better tell what I would do, I am not very experienced in iTunes.

I checked for third party plug ins and hand none. I'm set to 64 khz 24 bit audio. Still having the problem. It is a scratch muffled sound.

Unless you are not a musician/studio professional, you don't need 64Khz, 24 bit audio. Try to lower it to 44.1 Khz, 16bit 2 channels. That is the widely adopted "CD Audio" standard.

You already hear that sampling via system upsampling, sadly there is no 24bit consumer content around now (SASCD etc requires deck/hardware, has nothing to do with computer)

If you still get same problem and you are sure you have NOTHING tied to iTunes, e.g. plugin, enhancer,ever those Growl stuff, you must file a bug report to http://bugreporter.apple.com with all details they want with guidelines.
