The two drugs have a powerful effect on obese-mouse Insulin Resistance
(as do Rezulin, Actos and Avandia). That's half the problem for many Type
2.
What's exciting about this study, if it pans out in humans. . .and for
longer than 3 months, is that it attacks Insulin Resistance from a
different direction than either Actos/Avandia or Metformin. That means the
docs can add one or both of these to a TZD/Metformin regime and get more
anti-Insulin Resistance activity. Or, it offers another choice for
those who suffer too many side effects from the current anti-I.R. drugs.
But remember, they only attack Insulin Resistance, not the beta cell
death mechanisms which cause diabetes. I suggest that they won't handle
the excessive liver dump problem either.
My take on the article:
1. Obesity causes excess Free Fatty Acids (FFA)
2. FFA cause Insulin Resistance in cells
3. These two drugs operate by directly negating the FFA effect on
cells.
4. In contrast, Actos and Avandia work, in essence, by reducing FFA
in the blood.
5. Metformin uses two non-FFA attack mechanisms
That means the docs might develop a 1-2-3 attack instead of the current 1-2
attack.
Another interesting way to look at them:
1. High Insulin Resistance causes premature heart attack
2. High Insulin Resistance is common in non-diabetics
3. These two drugs could be low-side effect, anti-heart attack drugs
for non-diabetics
