I think there's a key distinction here. IMHO, JKR says that
"blood" does matter a great deal when you're talking about
someone's immediate family and how they relate to each other
(particularly how they love each other),
I agree, though I would change the causality so that the bond of blood becomes important /because/ of the love between them.
The basis for the strength of Dumbledore's protection of Harry (the one that relies on Aunt Petunia) was the protection Lily had alreday provided, which relied on Lily's sacrifice and her love for Harry. That protection, then, allowed Dumbledore to strengthen the protection he was adding by using the bond of blood between Lily and Petunia.
